RESUMO

Essential oils (EOs), including those from the Asteraceae plants, have been shown to have promising insecticidal activity against a wide range of insect pests. Understanding the mechanism of action of EOs is one of the studied aspects. The present study aimed to evaluate the effect of essential oils from Achillea millefolium, Santolina chamaecyparissus, Tagetes patula and Tanacetum vulgare on the settling and probing behavior of the bird cherry-oat aphid (Rhopalosiphum padi L.). In addition, the effect of the oils on the activity of such enzymes as trypsin, pepsin and α- and ß-glucosidase involved in the metabolism of proteins and sugars of the insects was examined. The leaf-choice bioassays demonstrated that the studied EOs limited aphid settling for at least 24 h after the treatment. The application of EOs also inferred with aphid probing behavior by reducing the total probing time and total duration of phloem sap ingestion. Aphids spent more time in the search phase due to an increase in the number and total duration of pathway phases. Moreover, the activity of the studied proteases and glucosidases significantly decreased in R. padi females exposed to the EOs. The enzyme inhibition varied depending on the applied oil and exposure time. Generally, the EOs with stronger deterrent activity also showed higher inhibitory effects. The results suggest that the tested EOs disrupt key digestive processes in R. padi which may be an important factor determining their aphicidal activity.

Changes in the level of oxidative stress markers-superoxide anion radical (O2-), hydrogen peroxide (H2O2) and malondialdehyde (MDA) and the activity of antioxidant enzymes-superoxide dismutase (SOD), catalase (CAT) and ascorbate peroxidase (APX) in the black bean aphid occurring on the primary host (viburnum plants) were studied. Among the aphid morphs, the lowest contents of O2-, H2O2 and MDA were noted for winged adults (alatae), which were also characterized by the highest activity of antioxidant enzymes. These metabolic features indicate the adaptation of winged morphs to the colonization of new host plants. During spring migration, an increase in the content of oxidative stress markers and antioxidant enzyme activities in wingless females (fundatrigeniae) was observed. The significance of the biochemical adaptation of the black bean aphid to its winter host is discussed.

Three aphid species, Eriosoma ulmi (L.), Colopha compressa (Koch) and Tetraneura ulmi (L.) induce distinct gall morphotypes on Ulmus glabra Huds.; opened and closed galls. Because the trophic relationship of aphids and their galls shows that throughout the gall formation aphids can elicit multiple physiological regulations, we evaluated the changes of hydrogen peroxide content (H2O2), cytoplasmic membrane condition, expressed as electrolyte leakage (EL) and concentration of thiobarbituric acid reactive substances (TBARS), as well as, the activity of catalase (CAT), guaiacol peroxidase (GPX) and ascorbate peroxidase (APX) in gall tissues, as well as, in damaged and undamaged parts of galled leaves. All aphid species increased EL from gall tissues and significantly upregulated APX activity in galls and galled leaves. Alterations in H2O2 and TBARS concentrations, as well as GPX and CAT activities, were aphid- and tissue-dependent. The development of pseudo- and closed galls on elm leaves did not have a clear effect on the direction and intensity of the host plant's physiological response. The different modes of changes in H2O2, TBARS, CAT and GPX were found in true galls of C. compressa and T. ulmi. Generally, physiological alterations in new plant tissues were quite different compared to other tissues and could be considered beneficial to galling aphids.

Thioredoxins (Trxs) and thioredoxin reductases (TrxRs) encompass a highly complex network involved in sustaining thiol-based redox homeostasis in plant tissues. The purpose of the study was to gain a new insight into transcriptional reprogramming of the several genes involved in functioning of Trx/TrxR system in maize (Zea mays L.) seedlings, exposed to the bird cherry-oat aphid (Rhopalosiphum padi L.) or the rose-grass aphid (Metopolophium dirhodum Walk.) infestation. The biotests were performed on two maize genotypes (susceptible Zlota Karlowa and relatively resistant Waza). The application of real-time qRT-PCR technique allowed to identify a molecular mechanism triggered in more resistant maize plants, linked to upregulation of thioredoxins-encoding genes (Trx-f, Trx-h, Trx-m, Trx-x) and thioredoxin reductase genes (Ftr1, Trxr2). Significant enhancement of TrxR activity in aphid-infested Waza seedlings was also demonstrated. Furthermore, we used an electrical penetration graph (EPG) recordings of M. dirhodum stylet activities in seedlings of the two studied maize varieties. Duration of phloem phase (E1 and E2 models) of rose-grass aphids was about three times longer while feeding in Waza plants, compared to Zlota Karlowa cv. The role of activation of Trx/TrxR system in maintaining redox balance and counteracting oxidative-induced damages of macromolecules in aphid-stressed maize plants is discussed.

Methicillin-resistant Staphylococcus aureus (MRSA) are among the most important biofilm-forming pathogens responsible for hard-to-treat infections. Looking for alternatives to antibiotics that prevent biofilm formation, we investigated the effects of manuka honey on the transcriptional profile of genes essential for staphylococcal biofilm formation using qRT-PCR. mRNA from two hospital MRSA strains (strong and weak biofilm producer) were isolated after 4, 8, 12 and 24 h from cells grown in biofilm. Manuka honey at 1/2 minimum biofilm inhibition concentration (MBIC) significantly reduced MRSA cell viability in biofilm. Manuka honey downregulated the genes encoding laminin- (eno), elastin- (ebps) and fibrinogen binding protein (fib), and icaA and icaD involved in biosynthesis of polysaccharide intercellular adhesin in both weakly and strongly adhering strain compared to the control (untreated biofilm). Expression levels of cna (collagen binding protein) and map/eap (extracellular adherence protein-Eap) were reduced in weakly adhering strain. The lowest expression of investigated genes was observed after 12 h of manuka honey treatment at 1/2 MBIC. This study showed that the previously unknown mechanism of manuka honey action involved inhibition of S. aureus adhesion due to reduction in expression of crucial genes associated with staphylococcal biofilm.

The effects of trans-cinnamaldehyde (TC) on transcriptional profiles of biofilm-associated genes and the metabolic activity of two methicillin-resistant Staphylococcus aureus (MRSA) strains showing a different degree of adherence to polystyrene, were evaluated. Metabolic activity of S. aureus in biofilm was significantly decreased in the presence of TC at 1/2 minimum biofilm inhibition concentration (MBIC). Expression levels of the genes encoding laminin binding protein (eno), elastin binding protein (ebps) and fibrinogen binding protein (fib) in the presence of TC at 1/2 MBIC were lower than in untreated biofilm in both the weakly and strongly adhering strain. The highest decrease of expression level was observed in case of fib in the strongly adhering strain, in which the amount of fib transcript was 10-fold lower compared to biofilm without TC. In the presence of TC at 1/2 MBIC after 3, 6, 8 and 12 h, the expression level of icaA and icaD, that are involved in the biosynthesis of polysaccharide intercellular adhesin, was above half lower in the weakly adhering strain compared to biofilm without TC. In the strongly adhering strain the highest decrease in expression of these genes was observed after 3 and 6 h. This study showed that TC is a promising anti-biofilm agent for use in MRSA biofilm-related infections.

The rose-grass aphid (Methopolophium dirhodum Walk.) is a major pest of maize (Zea mays L.), but little is known about the biochemical interactions between M. dirhodum and its host plant. Thiol compounds and glutathione S-transferase (GST) play a crucial role in the defense responses of maize to biotic stress factors, including aphids. The purpose of this research was to evaluate the impact of M. dirhodum herbivory on the total thiol (TT), protein bound thiol (PT), reduced glutathione (GSH) and oxidized glutathione (GSSG) contents as well as the activity of GST in three varieties of Z. mays (Zlota Karlowa, Ambrozja and Plomyk), that were classified as aphid-susceptible, aphid-relatively resistant and aphid-resistant, respectively. The earliest and strongest aphid-triggered alterations in the levels of TT, PT and GSH, and the greatest induction of GST activity, were recorded in the resistant Plomyk seedlings in relation to the relatively resistant Ambrozja and the susceptible Zlota Karlowa.

Prior experiments illustrated reactive oxygen species (ROS) overproduction in maize plants infested with bird-cherry-oat (Rhopalosiphum padi L.) aphids. However, there is no available data unveiling the impact of aphids feeding on oxidative damages of crucial macromolecules in maize tissues. Therefore, the purpose of the current study was to evaluate the scale of oxidative damages of genomic DNA, total RNA and mRNA, proteins, and lipids in seedling leaves of two maize genotypes (Zlota Karlowa and Waza cvs-susceptible and relatively resistant to the aphids, respectively). The content of oxidized guanosine residues (8-hydroxy-2'-deoxyguanosine; 8-OHdG) in genomic DNA, 8-hydroxyguanosine (8-OHG) in RNA molecules, protein carbonyl groups, total thiols (T-SH), protein-bound thiols (PB-SH), non-protein thiols (NP-SH), malondialdehyde (MDA) and electrolyte leakage (EL) levels in maze plants were determined. In addition, the electrical penetration graphs (EPG) technique was used to monitor and the aphid stylet positioning and feeding modes in the hosts. Maize seedlings were infested with 0 (control), 30 or 60 R. padi adult apterae per plant. Substantial increases in the levels of RNA, protein and lipid oxidation markers in response to aphid herbivory, but no significant oxidative damages of genomic DNA, were found. Alterations in the studied parameters were dependent on maize genotype, insect abundance and infestation time.

The major aim of the present study was to investigate the influence of juglone (JU; 5-hydroxy-1,4-naphthoquinone) treatments on the expression level of Cat1, Cat2 and Cat3 genes, encoding the respective catalase isozymes in maize (Zea mays L.) and wheat (Triticum aestivum L.) seeds. In parallel, germination efficiency, catalase (CAT) activity and hydrogen peroxide (H2O2) content in juglone-exposed cereal seeds were assessed. Juglone applications significantly stimulated abundance of three target catalase transcripts as well as induced CAT activity and generation of H2O2 in both maize and wheat kernels. Furthermore, germination process of juglone-affected maize seeds was more severe suppressed than in case of wheat kernels. The role of juglone in triggering the oxidative stress as well as antioxidative responses in seeds of the studied model cereal species are discussed.

The role of genes that are essential for development of Staphylococcus aureus biofilm during infection is not fully known. mRNA from two methicillin-resistant S. aureus strains that formed weak and strong biofilm on polystyrene plates were isolated at five time points from cells grown in biofilm and planktonic culture. Quantitative real-time PCR analysis showed that the expression levels of investigated genes under biofilm conditions were significantly higher than under planktonic conditions. The expression levels of the gene encoding elastin binding protein (ebps) and laminin binding protein (eno) were significantly increased in biofilm at 3 h, both in strongly and weakly adhering strain. The peak expression of fib gene encoding fibrinogen binding protein was found at 6 and 8 h in the case of strongly and weakly adhering strain, respectively. The expression of icaA and icaD genes in both strains was significantly higher under biofilm conditions when comparing to planktonic cells during 12 h. The expression level of the genes encoding binding proteins and the glucosamine polymer polysaccharide intercellular adhesin (PIA) slowly decreased after 24 h. Finally, we found that the expression levels of genes encoding binding factors in weakly adhering strain were significantly lower than in strongly adhering strain.

The developmental cycles of all B. burgdorferi s.l. genospecies present typical, main pattern described in the 90thies. The simple scheme might be modified according to the biology of species and hosts preference. Central European genospecies of B. burgdorferi s.l. can be associated with four groups of hosts playing the role of animal reservoirs. The group 1 contains genospecies associated with rodents as primary animal reservoir ­ B. afzelii, B. garinii, B. burgdorferi sensu stricto, strains B. bavariensis (B. garinii OspA serotype 4). The group 2 involves B. valaisiana and most of B. garinii strains, associated with birds. The group 3 involves B. spielmanii, the reservoir hosts are Gliridae, and hedgehogs. The group 4 includes B. lusitaniae, the hosts are lizards. B. miyamotoi enzootic cycle seems to be similar to B. burgdorferi complex, however, differs by the transovarial transmission possibility. The divisions are not extreme; in the hosts group, infected with appropriate Borrelia genospecies, very often are found the specimens infected with other genospecies.

In the Central European conditions, three species of Babesia have epidemiological significance as human pathogens ­ Babesia divergens, B. microti and B. venatorum. Tick Ixodes ricinus is considered as their main vector, wild mammals as the animal reservoir. The zoonotic cycles of small and large Babesia differ in details. Due to the lack of transovarial mode transmission in small species B. microti, the circulation goes mainly between immature ticks and vertebrate hosts; pathogen circulates primarily in the cycle: infected rodent → the tick larva → the nymph → the mammal reservoir →the larva of the tick. The tick stages able to effectively infect human are nymphs and adult females, males do not participate in the follow transmission. For large Babesia ­ B. divergens and B. venatorum, the transovarial and transstadial transmission enable the presence of the agent in adult ticks, moreover, that larvae and nymphs feed on not-susceptible hosts. The tick stages able to effectively infect cattle and other ruminants are adult females. Resuming, pathogen circulates primarily in the cycle the ruminant host ­ adult female tick ­ the larva ­ the nymph ­ adult female of the next generation ­ the ruminant. Due to the compound developmental transmission has place after the outflow of a tick began feeding.

One of the earliest responses of plants to insects' attack is generation of reactive oxygen species. However, the elevated level of ROS can elicit oxidative burst within plant tissues, and plants employ antioxidant systems against these radicals. Due to their chemical structures, polyphenols are able to diminish the level of ROS. Thus, we investigated the role of phenolic compounds in oxidative stress within winter triticale caused by Sitobion avenae and Oulema melanopus. It was found, that infestation by insects induced a high increase in the content of hydrogen peroxide and superoxide anion radical within resistant Lamberto cv. 24 hpi, whereas in sensitive Marko cv., an increase in H2O2 content was found within two days of aphid feeding. Moreover, resistant plants showed earlier and much greater induction of l-phenylalanine and l-tyrosine ammonia lyases and chalcone synthase activities, as well as accumulation of phenolic compounds in response to insect feeding than susceptible Marko. On the other hand, strong positive influence of hydrogen peroxide and superoxide radical contents on chalcone synthase activity and furthermore flavonoid biosynthesis was detected in the susceptible cultivar. Negative relationships between level of o-coumaric acid or flavonoid compounds and content of hydrogen peroxide or superoxide radical suggest their antioxidant capacity. Luteolin and o-coumaric acid may attend in scavenging of hydrogen peroxide, whereas quercetin, apigenin and (+)-catechin probably participate in reduction of superoxide anion radical content.

Plant NADPH oxidases (NOXs) encompass a group of membrane-bound enzymes participating in formation of reactive oxygen species (ROS) under physiological conditions as well as in response to environmental stressors. The purpose of the survey was to unveil the role of NADPH oxidase in pro-oxidative responses of maize (Zea mays L.) seedling leaves exposed to cereal aphids' infestation. The impact of apteral females of bird cherry-oat aphid (Rhopalosiphum padi L.) and grain aphid (Sitobion avenae F.) feeding on expression levels of all four NADPH oxidase genes (rbohA, rbohB, rbohC, rbohD) and total activity of NOX enzyme in maize plants were investigated. In addition, inhibitory effect of diphenylene iodonium (DPI) pre-treatment on NOX activity and hydrogen peroxide content in aphid-stressed maize seedlings was studied. Leaf infestation biotests were accomplished on 14-day-old seedlings representing two aphid-resistant varieties (Ambrozja and Waza) and two aphid-susceptible ones (Tasty Sweet and Zlota Karlowa). Insects' attack led to profound upregulation of rbohA and rbohD genes in tested host plants, lower elevations were noted in level of rbohB mRNA, whereas abundance of rbohC transcript was not significantly altered. It was uncovered aphid-induced enhancement of NOX activity in examined plants. Higher increases in expression of all investigated rboh genes and activity of NADPH oxidase occurred in tissues of more resistant maize cultivars than in susceptible ones. Furthermore, DPI treatment resulted in strong reduction of NOX activity and H2O2 accumulation in aphid-infested Z. mays plants, thus evidencing circumstantial role of the enzyme in insect-elicited ROS generation.

Reduced forms of ascorbate (AsA) and glutathione (GSH) are among the most important non-enzymatic foliar antioxidants in maize (Zea mays L.). The survey was aimed to evaluate impact of bird cherry-oat aphid (Rhopalosiphum padi L.) or grain aphid (Sitobion avenae F.) herbivory on expression of genes related to ascorbate-glutathione (AsA-GSH) cycle in seedlings of six maize varieties (Ambrozja, Nana, Tasty Sweet, Touran, Waza, Zlota Karlowa), differing in resistance to the cereal aphids. Relative expression of sixteen maize genes encoding isoenzymes of ascorbate peroxidase (APX1, APX2, APX3, APX4, APX5, APX6, APX7), monodehydroascorbate reductase (MDHAR1, MDHAR2, MDHAR3, MDHAR4), dehydroascorbate reductase (DHAR1, DHAR2, DHAR3) and glutathione reductase (GR1, GR2) was quantified. Furthermore, effect of hemipterans' attack on activity of APX, MDHAR, DHAR and GR enzymes, and the content of reduced and oxidized ascorbate and glutathione in maize plants were assessed. Seedling leaves of more resistant Z. mays varieties responded higher elevations in abundance of target transcripts. In addition, earlier and stronger aphid-triggered changes in activity of APX, MDHAR, DHAR and GR enzymes, and greater modulations in amount of the analyzed antioxidative metabolites were detected in foliar tissues of highly resistant Ambrozja genotype in relation to susceptible Tasty Sweet plants.

In Central European conditions, two species of Anaplasmataceae have epidemiological significance ­ Candidatus Neoehrlichia micurensis and Anaplasma phagocytophilum. Tick Ixodes ricinus is considered as their main vector, wild mammals as the animal reservoir. There is presented the transstadial transmission in ticks, due to the lack of transovarial mode the circulation goes mainly between immature ticks and hosts; pathogen circulates primarily in the cycle: infected rodent → the tick larva → the nymph → the mammal reservoir → the larva of the tick. The tick stages able to effectively infect human are nymphs and adult females, males do not participate in the follow transmission. The summary of available data of different A. phagocytophilum strains associations with different hosts revealed at least few distinct enzootic cycle, concern the same ticks species and different mammal hosts. It is possible to reveal in Central Europe the existence of at least three different epidemiological transmission cycles of A. phagocytophilum. The first cycle involves strains pathogenic for human and identical strains from horses, dogs, cats, wild boars, hedgehogs, possibly red foxes. The second cycle involves deer, European bison and possibly domestic ruminants. The third cycle contains strains from voles, shrew and possibly Apodemus mice. In Western Europe voles might be involved in separate enzootic cycle with Ixodes trianguliceps as the vector.

The north-eastern part of Poland is considered an area of high risk for infection with tick-borne diseases, including with human granulocytic ehrlichiosis (HGE) agents. The etiological agent of HGE is Anaplasma phagocytophilum. As the animal reservoir for A. phagocytophilum in the environment serve the species from Cervidae and Bovidae families. European bison (Bison bonasus) and elk (Alces alces) are the big ruminant species, reintroduced to the forests of Middle Europe after many decades of absence. In the foci of zoonotic diseases they are able to play a role as natural reservoir to pathogens, however, their status as protected animals means their study has been rare and fragmentary. The studies of B. bonasus were conducted in Bialowieza Primeval Forest and A. alces in Biebrza National Park. PCR amplifications were performed using primers amplifing the end of the groES gene, the intergenic spacer and approximately two-thirds of the groEL gene in the first round, and primers that span a 395-bp region of the groEL gene were used in the second round. The positive results were obtained in B. bonasus and A. alces, the prevalence of infection was 66.7 and 20.0%, respectively. Randomly selected samples were sequenced, sequences were compared with GenBank entries using Blast N2.2.13 and determined as A. phagocytophilum. The results presented herein are the first record of the presence of Anaplasma phagocytophilum in A. alces, and at the same time confirm the previous observations regarding the infection of B. bonasus with A. phagocytophilum.

The purpose of the study was to assess the prevalence and coinfection rates of Borrelia burgdorferi sensu lato genotypes in Ixodes ricinus (L.) ticks sampled from diverse localities in central and eastern regions of Poland. In years 2009-2011, questing nymphs and adults of I. ricinus were collected using a flagging method at 18 localities representing distinct ecosystem types: urban green areas, suburban forests and rural woodlands. Molecular detection of B. burgdorferi s.l. genospecies was based on amplification of a fla gene using nested PCR technique, subsequent PCR-RFLP analysis and bidirectional sequencing. It was revealed that 45 samples (2.1%) harboured two different B. burgdorferi s.l. genospecies, whereas triple infections with various spirochetes was found in 11 (0.5%) individuals. Generally, the highest average coinfection rates were evidenced in arachnids gathered at rural woodlands, intermediate at suburban forests, while the lowest were recorded at urban green areas. Overall, single spirochete infections were noted in 16.3% (n = 352/2,153) ticks. Importantly, it is the first report evidencing the occurrence of Borrelia miyamotoi (0.3%, n = 7/2153) in I. ricinus populations within central Poland. Circumstantial variability of B. burgdorferi s.l. genospecies in the common tick individuals sampled at various habitat types in central and eastern Poland was displayed. The coexistence of two or three different spirochete genospecies in single adult ticks, as well as the presence of B. miyamotoi were demonstrated. Therefore, further studies uncovering the co-circulation of the tested bacteria and other human pathogens in I. ricinus ticks are required.
